Formula
Round n0 up to whole people · p and d are fractions from 0 to 1 · z is dimensionless
| Symbol | Meaning | Unit |
|---|---|---|
| Required sample size before finite-population correction | people | |
| Standard-normal quantile for the chosen confidence level | dimensionless | |
| Expected population proportion; use 0.5 conservatively if unknown | fraction | |
| Desired half-width of the approximate interval | fraction |
n_0=\frac{z^2p(1-p)}{d^2}The formula follows from the normal approximation to the standard error of a proportion and assumes simple random sampling, sufficiently large n and a prespecified margin. A finite-population correction can reduce n; clustering, weighting, nonresponse and power for group comparisons require different planning. If p is unknown, p(1−p) is largest at p=0.5, producing the conservative largest sample size. This is a planning starting point, not a guarantee of precision under biased selection.
A representative simple random sample and normal approximation for one proportion are reasonable.
The desired half-width is translated into sample size through the standard error of a proportion.
Rearrange for margin
Approximate under simple random sampling.
95% confidence, p=0.30, d=0.05. Find n.
n0=1.962·0.30·0.70/0.052=322.7; at least 323.
Rounding down or confusing one-proportion estimation with a two-group difference design.
Round up and separately account for design, nonresponse, finite population and power needs.
Formula cluster
a smaller d raises required size quadratically.
For 95% confidence, z=1.96. With expected p=0.30 and half-width d=0.05, n0=1.962·0.30·0.70/0.052=322.7, so recruit at least 323 people. Check: using p=0.50 requires 385, more because it maximizes the variance. Add an allowance for nonresponse.
Plan representative prevalence and proportion surveys before data collection
Here is how to work through a typical Sample Size for Estimating a Proportion (n₀ = z² p(1−p) / d²) task step by step:
Task
95% confidence, p=0.30, d=0.05. Find n.
Solution path
n0=1.962·0.30·0.70/0.052=322.7; at least 323.
the sample size targeting a chosen margin for one proportion. Its output is a minimum count before design and nonresponse adjustments.. a smaller d raises required size quadratically. The quantity answers a specific biological or statistical question, not automatically the full cause of an observed pattern. Tie it to the population, sample or model level from which inputs came. A number without that reference can be misleading across studies. Keep description separate from explanation: the metric summarizes data but does not by itself establish the underlying mechanism.
Use z sets confidence, p expected variability and d the interval half-width. The units are p and d enter as proportions; n is rounded up to a whole person.. Check that numerator and denominator refer to the same time interval, target population and scale. Proportions entered as percentages usually need conversion to values between zero and one. Write units and definitions beside the observations. This helps catch swapped groups, mismatched age intervals or a wrong denominator before they distort the result. Also document measurement definitions and preprocessing, because studies can operationalize the same symbols differently.
The central assumption is: sampling is simple random and the goal is prevalence estimation rather than group comparison. p comes from prior evidence or conservatively uses p=0.5. If clustering, weighting or nonresponse reduce effective information., interpretation changes or the formula no longer fits. State sampling, measurement method and biological scale. A model is useful because it simplifies a question, but its limits belong in the report. Do not automatically transfer parameters to different species, populations, laboratories or sampling conditions. Report these assumptions with the result so readers can judge which populations and conditions the conclusion can cover.
In the example, z=1.96,p=0.30,d=0.05 require at least 323 people. The check is: p=0.5 has greater variance and requires 385 cases under the same settings. Substitute calculated values back into the original relation and check whether ranges and units are biologically plausible. Rounded data can create a small discrepancy, but should not change direction or order of magnitude. Also test a simple limiting case, such as zero, complete detection or equal groups. This separates arithmetic errors from model limitations. Round intermediate values only at the end, and check the order of magnitude against the biological question.
A nearby quantity is sample-size planning for comparing two means or proportions. The substantive distinction is that this plans precision for one proportion; group comparisons also need effect size and power. Both can appear in the same biological analysis, but they use different inputs and answer different questions. Choose by the requested output and verify the data definition before comparing values. A related name does not imply identical units or interchangeable interpretation. This matters when a descriptive statistic is reported alongside a model parameter. Before comparing values, align definitions, populations, observation periods and analysis rules; otherwise the difference may be methodological.
